i'm not sure if I understand your question correctly. basically, the circular DNA will not come out of the plug. the band at 1.25mb is the linearized genomic. the marker is also made up of linear fragments. Large circular fragments do not enter the PFGE gel.
thus, the linearized genomic DNA can be separated from the circular by PFGE. in that experiment, they ran the gel for awhile so that all the linearized fragments will leave the plug and enter the gel while the circular genomic DNA (which is they want will stay behind).

Silver service is very easy to pick up. You would probably be able to learn on the job, it should be part of the training.
Semi Silver service is when you have to serve things up at the table from a platter. Usually the chef will carve the meat (or whatever) in the kitchen, then you would take the plates to the table, you would then hold a spoon and a fork in one hand and serve up potatoes and vegetables. If you wanted to practice then hold the fork between your index finger and thumb and hold the spoon with your other three fingers so that you can scoop with the spoon and hold things on the spoon with the fork (If this makes sense…)
Silver service is exactly the same principle as semi silver but you have to serve everything at the table, including any meat/fish etc..
Normal plate waiting is when the whole meal is plated up in the kitchen and you deliver it to the table or, the meat/fish etc is put on the plate by the chef and you deliver potatoes and veg in a side dish.

There are several issues to be taken into account when using older lenses on a new digital SLR. First is that you will lose the autofocus and/or auto aperture features of your camera, also you will have to take into account the size difference between the CMOS sensor in your camera and a 35 mm frame. The sensor being smaller will mean that you will have to make some calculations as to what the actual zoom on a given lens will be. For example if you have a 55mm lens for a 35mm camera, it will actually function as a ~ 82.5mm lens (you would use the actual mm rating and multiply it by 1.5).

There really arent too many general purpose lenses that are a while lot better optically for the same price. You can look into some third party lenses like the Tamron 17-50 f/2.8 or Sigma 24-70 f/2.8. They are all decent, but if you dont need the f/2.8 dont waste your money.
